In Vitro Cultivation of Early Rat Embryos1

In vitro cultivation of early mammalian embryos under controlled con ditions in various kinds of media have been reported by numerous investigators. According to Austin's detailed review in 1961, the majority of studies in this field have involved the rabbit, mouse, and a few other mammalian species. It has been shown that rabbit ova will cleave readily and develop into morulae in a defined medium supplemented with a large amount of serum, although the expansion of the blastocyst is still refractory. Mouse ova can be cultured from the two-cell stage to full expanded blastocysts, but until 1962 the first division was still refractory (Brinster 1963, 1965a, 1965b, 1965c), Gwatkin (1963), Mintz (1964). In that year, Biggers et al. used the organ culture method with mouse Fallopian tubes, and cultured ova successfully from the one-cell stage through blastocyst. They noted that under these conditions, much of the cellular integrity of the tubes was preserved normally. Rat ova, however, have continued to defy such attempts and, at present, they can not be cultured successfully in vitro. The present investigation was undertaken to explore various methods of in vitro cultivation of early rat embryos.
